My favorite bit (and this is going to sound dumb) was actually Sam's line after Dean says he's going to go scrape some hearts of the barroom floor--"You go be you", with absolutely not judgement in his voice at all, just fond affection. Early show Sam actually would have been "judgy", but even in the kitchen scene at the beginning, he really wasn't--just amused, and teasing because that's what brothers do. (Same thing back in "Baby"; I really like how they've reached this point where each pretty much accepts who the other is. Sure, Dean is baffled by Sam's preferences a lot of the time, but more in the "I just don't understand why you would/wouldn't want to do that, but okay--guess I'll go do it by myself (but are you sure you don't want to come?)" -way, not "you're a kill-joy" -way.)
In other words, this episode was just so awesome, and this season as a whole has been awesome, and I'm lying on the floor in a puddle of show-love right now. It's pretty glorious.
